Further information: | The plasmid was constructed by PCR amplifying the uracil-specific excision reaction (USER) cassette, ccdB gene and chloramphenicol resistance gene from pCfB49 and ligating the fragment into pESC-URA. Finally, the ccdB gene and chloramphenicol resistance gene were removed to obtain pCfB132. The plasmid contains a S. cerevisiae 2μ FLP Recombinase Target site (FRT site), next to the USER cloning cassette (AsiSI and Nb.BsmI). The plasmid is an episomal replication vector intended for USER cloning and USER fusion. The nucleotide sequence of the parental pESC-URA vector corresponds with the EMBL Nucleotide Sequence Database accession number AF063585.2. Other name of the plasmid is pESC-URA-USER. |
