| Further information: | The plasmid was constructed as follows: 1) a cDNA library from a Nicotiana plumbaginifolia var. P2 (tobacco) cell suspension culture was constructed in pUC18; 2) an oligo of 29 nucleotides, matching the N-terminal part of the coding sequence of the tobacco mitochondrial manganese superoxide dismutase cDNA (MnSOD) was used as a probe to screen; 3) the clone with the largest cDNA insert was selected. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727069.1. The nucleotide sequence of the MnSOD cDNA corresponds with the Genbank accession number X14482.1. |
