Further information: The λN22 protein recognises a 15 nt RNA stem-loop (box-B) with high specificity. Fused to the fluorescent mVenus tag, this system can be used to visualise the localisation of RNAs in plant cells.
The plasmid contains the nptII gene which allows selection on neomycin in plant cells.
Target RNAs can be introduced by Gateway recombination with p35S:16xBoxB-Gateway-BASTA (LMBP 9017).
The λN22/box-B system is well suited for low-background and high-throughput studies.
When compared with EYFP, mVenus contains the following mutations: F46L, F64L, M153T, V163A and S175G.
Other names of the plasmid are pSCJ383 and pUbiQ:IN22-mVenus-NLS NPTII.
