Further information: | The plasmid was constructed as follows: a BamHI fragment from pUR250IVHA8, coding for secreted heamagglutinin (the last 36 amino acids are missing) was inserted into the unique BamHI site of pUR250. The orientation of the inserted fragment is such, that the HA8s gene can be cloned into expression vectors as BamHI or as SalI fragments. This haemagglutinin coding sequence is also called anchor-free haemagglutinin (HA(-)); it lacks the sequences coding for the intramembranous part and the cytoplasmic tail of the protein. This plasmid is a basic construction for all HA8s containing vectors. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. Other names of the plasmid are pUR250H8A- and pUR250HA8s. |
