Further information: The plasmid was constructed by preparing the human ACLY (Uniprot ID P53396-2) coding sequence from poly A+ RNA from human liver (Clontech) and cloning it into the pTrcHis2-TOPO vector, in frame with the C-terminal Myc epitope and His-tag.
The human ACLY coding sequence corresponds with isoform 2, which differs from the canonical sequence in the fact that it is shorter at the N-terminus and is lacking an internal segment.
The nucleotide sequence of the human ACLY coding sequence corresponds with the Genbank accession number NM_198830.1.
Other names of the plasmid are pTRcHis2-hACL_full_length, pTrc-His2-hACL and pTRcHis2-hACL_WT.
