| Recipe: | Casitone 10,0 g Yeast extract 2,5 g Glucose 2 g Cellobiose 2 g Soluble starch 2 g Clarified rumen fluid 150-300 ml KH2PO4 0,45 g K2HPO4 0,45 g (NH4)2SO4 0,9 g NaCl 0,9 g MgSO4 x 7H2O 0,09 g CaCl2 0,09 g Cysteine-HCl 1,0 g NaHCO3 4 g Reazurin 1 mg Distilled water 1 L Mix ingredients, except NaHCO3 and L-cysteine HCl, and adjust pH to 6. Boil until a red color develops. Dispense the medium into suitable culture vessels under a stream of H2-CO2 and seal with butyl rubber stoppers. Autoclave. Prior to inoculation, aseptically and anaerobically add the filter sterilized L-cysteine•HCl and NaHCO3 solutions. |
