Accession number: LMBP 130
Depositor: Prof. E. Remaut1 2
1 Department for Molecular Biomedical Research, VIB, Ghent, Belgium
2 Department of Biomedical Molecular Biology, Ghent University, Ghent, Belgium
Containment level: This strain has been assigned the containment level Class 1 following the European Directive 2009/41/EC on the contained use of genetically modified organisms, and its updates (see also the Belgian risk group classification).
Medium: LB-Lennox
Selection marker: /
Temperature: 28°C
Genotype: lacZam trpAam rpsL λ(bio252 cI857 ΔH1)
Phenotype: StrR
Properties: Expression host for plasmids containing phage λ's PL or PR promoter. Induction is obtained by shifting the culture from 28°C to 42°C. The resident λ is a defective lysogen. ΔH1 removes part of cro and all genes to the right of it. bio252 removes all genes to the left of cIII. cI857 is ind-. At 42°C, N is expressed from the lysogen. As a consequence, transcription termination signals that might be present downstream from the PL promoter and the nutL site, as present on the pPL-series of plasmids, can be overridden.
Further information: Useful for the expression of cloned genes carrying transcription terminators between the start of the gene and the insertion point.
