1 Experimental Biophysics, Institute of Biology, Humboldt University of Berlin, Berlin, Germany
Containment level: This strain has been assigned the containment level Class 1 following the European Directive 2009/41/EC on the contained use of genetically modified organisms, and its updates (see also the Belgian risk group classification).
Other culture collection numbers: c043
Medium: LB-Lennox
Selection marker: /
Temperature: 28°C
Genotype: F- ompT gal dcm hsdSB (rB- mB-) (DE3) manX::ribM ribC::cat-ribF glnA- asnB-
Phenotype: Riboflavin and glutamine auxotrophy
Properties: The strain is intended for amino acid specific labelled protein production and flavin labelling. As a C41(DE3) derivative, this strain contains DE3, a λ prophage carrying the T7 RNA polymerase gene under control of the lacUV5 promoter and lacIq. IPTG is required to induce expression of the T7 RNA polymerase. Protein expression from transforming plasmids containing a T7 promoter-driven expression system is repressed until IPTG induction of T7 RNA polymerase expression occurs. The strain is deficient in the outer membrane protease OmpT which reduces the degradation of heterologous proteins expressed in the strain.
Further information: This strain was created based on E. coli B CpXribF (LMBP 13316) via site specific chromosomal modification.
