Further information: The plasmid was constructed by cloning the BglII/NcoI fragment from pAN52-1, containing the gdpA promoter, into the BglII/NcoI opened pHSP70-SG vector, upstream of the SGFP coding sequence. The gpdA-hyg-trpC cassette from pAN701 was excised as a BglII/BamHI fragment and cloned into the BglII site of the newly created vector.
The nucleotide sequence of the gdpA promoter corresponds with the EMBL Nucleotide Sequence Database accession number M33539.1.
The nucleotide sequence of the trpC terminator corresponds with the EMBL Nucleotide Sequence Database accession number X02390.1.
Since the nucleotide sequence of the gGFP vector could not be completely traced back, there is uncertainty as to the cutting frequency of the restriction enzymes, except for the sites that were analysed in the authenticity test.
