Further information: | The plasmid was constructed by cloning a 5 kb EcoRI-SalI fragment from the Charon 4A gene bank, containing the acetamidase gene from A. nidulans, into the unique EcoRI and unique SalI site of pBR322. The nucleotide sequence of the amdS gene corresponds with the EMBL Nucleotide Sequence Database accession number M16371.1. Since the nucleotide sequence at the 5' and 3' end of the insert could not be completely traced back, there is uncertainty as to the cutting frequency of the restriction enzymes, except for the sites that were analysed in the authenticity test. |

Authenticity test: | Restriction enzyme pattern analysed at GeneCorner: AvaI, BsaAI, EcoRI, EcoRI/SalI, EcoRI/XcmI and XcmI. The results 1) EcoRI, SalI and XcmI are really unique sites; 2) The plasmid length seems to be approx. 9000 bp. The approximately 200 extra nucleotides are located between the EcoRI site and the Xcm site, most probably in the unknown sequence region; 3) The orientation of the acetamidase insert could be confirmed; 4) An extra AvaI site and an extra BsaAI site have been found in the acetamidase terminator region. |
