Further information: The plasmid was constructed by substituting the HindIII-XbaI fragment from p415GALL by the HindIII-XbaI fragment from pSCGAL1mBAX-L, containing the α variant of the mouse Bcl2-associated X cDNA (mBAX).
p415GALLmBAX is a low-copy number shuttle vector for expression of the mouse Bax-α protein.
The GALL promoter is a deletion variant of the Saccharomyces cerevisiae galactokinase promoter (GAL1), which lacks one of the three UAS elements required for full induction of the promoter by galactose. The GALL promoter is tightly repressed by glucose and is induced at a moderate level by galactose.
When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
The nucleotide sequence of the mBAX cDNA corresponds with the EMBL Nucleotide Sequence Database accession number L22472.1.
