Further information: The plasmid was created as follows: 1) the attP1r sites was isolated as a NotI/SalI fragment from pDONR-P4-P1R and cloned into the NotI/SalI opened pBluescriptKS+ vector, generating pBS-P1R, 2) the attP4 sites was isolated as a ApaI/XmnI fragment from pDONR-P4-P1R and cloned into the SmaI-opened pBluescriptKS+ vector, generating pBS-P4, 3) the hygromycin resistance cassette was isolated as a HpaI fragment from pDONR-A-Hyg and cloned into the SmaI-opened pBluescriptKS+, generating pBS-A-Hyg 4) the hygromycin resistance cassette was isolated as a EcoRI/NotI fragment from pBS-A-Hyg and cloned into the EcoRI/NotI-opened pBS-P1R, generating pBS-P1R-A-Hyg and 5) the attP4 site was isolated as an EcoRI/SacI fragment from pBS-P4, the fragment was blunted and cloned into the NotI-opened and blunted pBS-P1R-A-Hyg, generating pA-HYG OSCAR.
Name used in Paz et al (2011) is pA-Hyg-OSCAR.
The nucleotide sequence of the pOSCAR vector corresponds with the EMBL Nucleotide Sequence Database accession number HM623915.1.
