Further information: The plasmid was constructed via allelic exchange between pYV40 and pPW52, containing a mutant version of Y. enterocolitica yopE, designated yopE21.
Y. enterocolitica yopE21 has a translational frame-shift following a 4 bp insert between codons 3 - 4. An additional BglII site is present at this place.
This plasmid was the first step towards the ΔHOPEMT multimutant strain (pYV40 yopE21 yopHΔ1-352 yopOΔ65-558 yopP23 yopM23 yopT135).
Other names of the plasmid are pYV40 yopE and pYV40 yopE21.
