Further information: This plasmid was constructed as follows: pCaMET15 was cleaved with BanII (blunted with T4 DNA polymerase)/AatII and the resulting 4044 bp fragment (containing part of the Candida albicans O-acetylhomoserine O-acetylserine sulfhydrylase (CaMET15) genomic DNA, the origin of replication and the ampicillin resistance gene) was ligated into the XmnI/AatII opened pACTyEGFPTU vector.
The orientation of the MET15 coding sequence is sense relative to the URA3 gene.
pACT1EGFPTUMD1 is a stable integrative vector and allows integration of the linearized vector into the Candida albicans genome via homologous recombination at the MET15 locus.
The origin of the sequence between the MAL2 terminator and the yEGFP3 sequence is unknown.
The nucleotide sequence of the CaMET15 genomic DNA corresponds with the EMBL Nucleotide Sequence Database accession number AF188275.1.
The nucleotide sequence of the CaURA3 DNA corresponds with the EMBL Nucleotide Sequence Database accession number X14198.1. HOWEVER, THERE MIGHT BE CORRESPONDENCE WITH AF109400.1: see LMBP 4100.
