Further information: | The plasmid was constructed by ligating a 359 bp fragment of the E35S promoter/plant junction region into the pCRII vector with the TA cloning Kit Dual Promoter (Invitrogen). The cloned DNA consists of a junction between part of the 35S promoter from the Cauliflower Mosaic virus (CaMV) and plant DNA, as present in the transgenic Roundup Ready (RR) soybean line event 40-3-2 (Monsanto). The CaMV promoter fragment is used as a promoter gene in this transgenic event. The E35S promoter/plant junction is highly specific for RR soybean and admits unambiguous identification of the genetically modified organism. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727196.1. The nucleotide sequence of the inserted E35S promoter/plant junction region fragment corresponds with the EMBL Nucleotide Sequence Database (Release 67; Accession number AJ308514). |
