Further information: Derived from pAT153 by inserting the human immunoglobulin γ3 gene as an EcoRI-HindIII fragment. This insert, which was isolated from the partial HaeIII-AluI Charon 4A genomic library (Maniatis), encodes the constant portion of the human γ3 heavy chain only.
In B-cell differentiation, the 5' flanking region of the hIG3 gene is removed by joining to the J region.
Since part of the nucleotide sequence of the 3' flanking region of the hIG3 insert (between nucleotide position 4 and 1113 in pATHIG31) is unknown, there is uncertainty as to the cutting frequency of the restriction enzymes indicated.
The nucleotide sequence of the hIG3 sequence used for the computer enzyme map is taken from Huck et al. (1986) and some differences with our cloned hIG3 gene can be expected: e.g. 1) at nucleotide position 966 of hIG3 we found a C while an A is mentioned in Huck et al.; 2) at nucleotide position 1158 we found a G instead of the A mentioned by Huck et al. (1986).
