Genecorner:

-

Cloned DNA: -
Promoter: Human interleukin 6 promoter (IL6)
Ribosome
binding site:
-
Terminator: Simian virus 40 polyadenylation signal (SV40 polyA)
Selection marker: Ampicillin (amp)
Replicon: Escherichia coli plasmid pMB1 origin
Host range: Escherichia coli
Parental clone: pMaSVTHIL61; pSV23S; pAT153
Further information: The plasmid was constructed by a three fragment ligation: 1) the EcoRI-StuI fragment of pMaSVTHIL61, containing the human IL6 promoter; 2) the StuI-BamHI fragment of pSV23S, containing the SV40 polyA and the SV40 small t-antigen splicing signal; 3) the BamHI-EcoRI opened pAT153 vector. In this way, 5 unique sites (AvrII, HindIII, StuI, XbaI, XhoI) were obtained to clone a gene under control of the human IL6 promoter.
The tetracycline resistance gene was disrupted.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727532.1.
The nucleotide sequence of the genomic human IL6 DNA corresponds with the EMBL Nucleotide Sequence Database accession number AC073072.11, except for the C nucleotide at promoter position -174 (polymorphism mutant) which was changed into a G nucleotide (wild type).
Other name of the plasmid is pATIL6PpolyA.
