| Further information: | pATgMIFNG is a pAT153 vector with in the BamHI site a 10.5 kb BamHI fragment of the EMBL 3A-7Mug phage, containing the genomic mouse interferon γ gene (mIFNG). The orientation of the inserted gene is unknown. Since the nucleotide sequences of the introns of the genomic mIFNγ gene could not be completely traced back, there is uncertainty as to the cutting frequency of the restriction enzymes, except for the sites that were analysed in the authenticity test. The approximative length of the plasmid and of the mIFNG insert deduced from the compiled nucleotide sequence could be confirmed. Only the 1509 bp PvuI/BglII fragment, containing part of the 3' flanking region of the mIFNG gene, was clearly (+-) 200 bp smaller. Furthermore, the results revealed that EcoRI cuts at least 6 times. The location of the extra EcoRI sites have not been determined; consequently, the compiled nucleotide sequence was not adapted. Other name of the plasmid is pATgMIFNGB. |
