Further information: | pBARGPE1 is a fungal expression vector designed to express genes or cDNAs under their own initiation codons. The plasmid carries a lox-NotI-lox sequence for insertion into lambda and for site-specific recombination between lox sites, catalyzed by the Cre protein. The plasmid also carries the Streptomyces hygroscopicus bar gene conferring resistance to the herbicide bialaphos. The nucleotide sequence of the bar gene corresponds with the EMBL Nucleotide Sequence Database accession number X05822.1. The nucleotide sequence of the gdpA promoter corresponds with the EMBL Nucleotide Sequence Database accession number M33539.1. The nucleotide sequences of the trpC promoter and terminator correspond with the EMBL Nucleotide Sequence Database accession number X02390.1. Since the nucleotide sequence of the pBARGPE1 vector could not be completely traced back, there is uncertainty as to the cutting frequency of the restriction enzymes, except for the sites that were analysed in the authenticity test. |
