Further information: This cat reporter plasmid was constructed by deleting 467 bp of vector sequence immediately upstream of the polylinker from pBLCAT3 (Luckow et al., 1987), and replacing it by two fragments from SV40 DNA, inserted in tandem and each containing the SV40 polyadenylation signal.
pBLCAT6 contains the SV40 small t-antigen splicing signal.
This plasmid contains three SV40 polyA segments; hence, the possibility might exist that the plasmid could lose information by homologous recombination; so far, no observations have been made in this respect (Boshart, personal communication).
pBLCAT6 is most useful for insertion of an eukaryotic promoter sequence and determination of the corresponding expression efficiency under various conditions.
When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
