Further information: | The plasmid was constructed as follows : 1) After digesting pSV23SE6L with HindIII, the 5' sticky ends were filled in with Klenow polymerase and SalI linkers ('CGTCGACG') were ligated; 2) The SalI fragment, containing the gene encoding the light chain E6L of the mouse monoclonal antibody E6(IgG2b,κ), was then cloned into the unique SalI site of pBMGNeo, clockwise behind the mMt1 promoter. The mMt1 promoter controls the expression of the inserted E6L gene in mammalian cells. Because of the presence of the second intron of the rabbit β-globin gene in the 3' untranslated region, the E6L transcript is spliced. The 3' untranslated region of the E6L gene still contains 15 dC-nucleotides of the dG/dC tail derived from the cDNA synthesis of E6L (De Waele et al. (1988)). |
