Further information: | The plasmid was constructed by cloning the OMMB gene as a PstI fragment (nucleotide position 3645 and 4660) from pOMMB into the unique I-PstI site of pBR325. As compared to the complete OMMB gene, the leader signal fragment and 3 codons of the variable region fragment are removed in the OMMBf gene. The length of the dG/dC-tail after the 3' untranslated region is unknown. The nucleotide sequence encoding the human γ heavy chain disease (HCD) OMM protein was obtained from Genbank 'Release 55; Locus : HUMIGHAF'. The unknown nucleotides in the Genbank-obtained sequence have been sequenced (Dr R. Schoonjans and K. De Sutter, Department of Biomedical Molecular Biology, Ghent University, Belgium). OMMBf differs from the total OMMB gene in that the Lf (leader signal fragment) and 3 amino acids of VHf are omitted, due to the PstI site in VHf (nucleotide position 3645 in pOMMB), used in the cloning procedure. Other name of the plasmid is pBR325OMMB. |
