| Further information: | The plasmid was constructed by inserting an approx. 7 kb EcoRI fragment from λCH4A-gHIIF-2, containing part of the genomic human IFNγ gene and its 5' flanking region, into the unique EcoRI site of pBR328. The orientation of the inserted fragment is not known. The nucleotide sequence of the hIFNG gene is a concatenation of two entries obtained from the EMBL Nucleotide Sequence Database accession numbers M32746 and J00219, preceded by an unknown nucleotide sequence fragment of approx. 2440 bp. The nucleotide sequence of the signal sequence of the hIFNG gene is located from nucleotide 6803 up to and including 6871. As such, the mature hIFNγ is 3 amino acids shorter as compared to the amino acid sequence described in Taya et al. (1982). Since part of the nucleotide sequence of the 5' flanking region of the hIFNG gene is not known, there is uncertainty as to the cutting frequency of the restriction enzymes indicated. Other name of the plasmid is pBR328γE7.0. |
