|Further information:||The plasmid was constructed by inserting the KpnI-SalI fragment of pSV23SE6Hm (containing E6H gene, without the leader signal sequence) between the KpnI and SalI sites of pBRphoA. Both KpnI sites were previously blunted with T4 (exonuclease), which connects the fusion in phase.
The presence of the lacI(q) repressor gene is necessary to control the activity of the tac promoter.
