|Further information:||The plasmid was constructed by ligation of three fragments: 1) the large EcoRI-SalI fragment (containing ori and amp) of pBRphoAE6Hf2; 2) the EcoRI-ApaLI fragment (containing tac promoter, signal sequence of phoA, the variable domain and a fragment of the constant domain 1 of E6H) of pBRphoAE6Hf2; 3) the ApaLI-SalI fragment of pSV51cE6Hy3 (containing the completing part of constant domain 1, the hinge region and the constant domains 2 and 3 of hIG3).
The sphoA-E6Hy3 fusion is in phase.
The presence of the lacI(q) repressor gene is necessary to control the activity of the tac promoter.
