| Further information: | The plasmid was constructed by ligation of three fragments: 1) the large EcoRI-SalI fragment (containing ori and amp) of pBRphoAE6Hf2; 2) the EcoRI-ApaLI fragment (containing tac promoter, signal sequence of phoA, the variable domain and a fragment of the constant domain 1 of E6H) of pBRphoAE6Hf2; 3) the ApaLI-SalI fragment of pSV51cE6Hy3 (containing the completing part of constant domain 1, the hinge region and the constant domains 2 and 3 of hIG3). The sphoA-E6Hy3 fusion is in phase. The presence of the lacI(q) repressor gene is necessary to control the activity of the tac promoter. Name mentioned in Flamez et al. (1995) is pBRphoAcHy3H. |
