Further information: The plasmid was constructed by ligation of three fragments; the large EcoRI-SalI fragment (ori and amp) and the EcoRI-ApaLI fragment (tac promoter, signal sequence of phoA, the variable domain and a fragment of the constant domain of E6H) of pBRphoAE6Hf2, with the ApaLI-SalI fragment of pSV51cE6Hy3f2 (containing part of the constant domain 1, the hinge and constant domain 2 of the cDNA of hIG3).
This plasmid is similar to pBRsE6Hy3, but the cDNA encoding the mouse::human chimeric heavy chain of E6(Hy3,κ) is not complete. After translation, E6Hy3f2 contains the first 27 amino acids of the original CH2 domain followed by 4 amino acids derived from the cloning strategy and the multi-stop-linker.
The sphoA-E6Hy3f2 fusion is in-phase.
The presence of the lacI(q) repressor gene is necessary to control the activity of the tac promoter.
