Cloned DNA: Chloramphenicol acetyltransferase gene (cat)
RNA one modulator gene (rom)
Promoter: -
Ribosome
binding site:
-
Terminator: -
Selection marker: Chloramphenicol (cam)
Replicon: Enterobacter agglomerans plasmid pPIGDM1 origin
Host range: Escherichia coli
Enterobacter agglomerans
Parental clone: pPIGDM1; pBR328
Further information: This plasmid was constructed by inserting a HindII fragment of pBR328, containing the chloramphenicol resistance gene, into the unique HindII site of pPIGDM1.
pBW4 carries the rom gene, regulating the copy number of the plasmid (10 to 12 in E. coli K12 MG1655).
pBW4 can be used for the construction of cloning vectors compatible with ColE1/pMB1-, p15A-, RSF1030- or CloDF13-derived plasmids.
Dr B. Wrobel communicated the presence of an unexpected HindIII site near the HindII junction.
Sequence analysis, performed at LMBP, revealed the presence of an additional 432 bp HindII fragment of unknown origin, containing the unexpected HindIII site.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727305.1.
The nucleotide sequence of the pPIGDM1-derived part of the plasmid corresponds with the EMBL Nucleotide Sequence Database accession number AF014880.1.
The nucleotide sequence of the inserted HindII fragment of pBR328 corresponds with the EMBL Nucleotide Sequence Database accession number L08858.1.
