| Further information: | This plasmid is constructed by inserting a mouse GAPDH fragment (clone 2G61F from P19EC library of L. Jonk) provided with an EcoRI/SmaI adaptor (GAATTCCCGGG) in the unique EcoRI site of the polylinker of pBluescriptIIKS-. GAPDH is a 'house keeping' gene and can be used as hybridisation control. The inserted fragment is homologous with residue 115-519 of rat GAPDH cDNA. It contains 41 nucleotides of the 5' UTR region and the first 436 nucleotides of the mGADPH coding sequence. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. |
