Further information: The plasmid was created by inserting the SalI restriction fragment containing GFP into the SalI opened vector pUCSAM and selecting for sense oriented ligation product, resulting in a GFP-SAM fusion protein. The XmaI-SstII restriction fragment containing GFPSAM was ligated into the XmaI-SstII opened pCAGGS-SAMase vector.
The pCAGFP vector consists of the pCAGGS vector with the GFP sequence inserted.
This vector produces a fusion protein GFP-SAMase in which SAMase is active.
When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
