Further information: | The plasmid was constructed by removing the C-terminal fragment of human CYLD from pCAGGS-E-CYLD-C via EcoNI/NsiI restriction enzyme digest and replacing it with a mutated fragment obtained from HA-CYLD-C601A. The plasmid contains a human CYLD fragment in which the catalityc cysteine residue at position 601 was mutated to alanine. The construct was verified to express and was functionally validated by its inability to block TRAF2-induced NF-kappaB induction in a luciferase assay. The nucleotide sequence of the wild type human CYLD cDNA corresponds with the EMBL Nucleotide Sequence database accession numberAJ250014.1. |
