Further information: The plasmid was constructed by isolating an N-terminal fragment of the human CYLD coding sequence, containing the R324A mutation, from pCAGGS-E-CYLD-R324A as a HincII fragment and cloning it into the HincII opened pCAGGS-E-CYLD-C-C601A vector.
The human CYLD coding sequence contains an R324A mutation in the human MALT1 cleavage site and a C601A mutation in the catalytic site of the enzyme.
The plasmid was confirmed by the depositor on expression, MALT1 cleavage and lack of inhibition in luciferase assays.
The nucleotide sequence of the wild type human CYLD cDNA corresponds with the EMBL Nucleotide Sequence database accession number AJ250014.1.
