Further information: pCAGGS-E-exoS-GAPminADPRTmin is useful for highly efficient expression of mutated exoS, lacking GAP and ADPRT activity, under control of the AG promoter and the hCMV-IE enhancer in various mammalian cells.
As compared to the wt exoS cDNA, the arginine (R) codon at position 146 was replaced by a lysine (K) codon and the glutamic acid (E) codon at position 381 was replaced by an aspartic acid (D) codon.
When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
The nucleotide sequence of the wt exoS cDNA corresponds with the Genbank accession number NC_002516.2.
