Simian virus 40 bidirectional origin (SV40)
Host range: Escherichia coli
Mammalian cells; SV40 permissive cells
Parental clone: pAS2-hA20-C103S; pCAGGSEhA20
Further information: The plasmid was constructed by inserting the EcoNI-HpaI fragment from pAS2-hA20-C103S into the EcoNI-HpaI opened pCAGGSEhA20 vector.
pCAGGS-E-hA20-C103S is useful for efficient expression of mutated hA20 under the control of the AG promoter and the hCMV-IE enhancer in various mammalian cells.
The hA20 insert contains a mutation in the ovarian tumor (OTU) domain (C103S).
When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727012.1.
The nucleotide sequence of the wt hA20 cDNA corresponds with the EMBL Nucleotide Sequence Database accession number M59465.1.
Other name of the plasmid is pCAGGS-E-A20(C103S).
