Further information: | The plasmid was constructed as follows: 1) the human ubiquitin B mature sequence was PCR amplified on the template pET3E-His-AU1-hUb; 2) the KpnI and partially SalI digested amplicon was then inserted into the KpnI and SalI opened pCAGGS-FLAGmA20 plasmid. pCAGGS-FLAG-hUb is useful for highly efficient expression of human Ub under the control of the AG promoter and the human CMV-IE enhancer in various mammalian cells. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT726814.1. The nucleotide sequence of the human Ub insert, which was the result of the sequence analysis at BCCM/GeneCorner, differs from the nucleotide sequence obtained from the EMBL Nucleotide Sequence Database accession number BT020104.1 in some silent mutations. Other name of the plasmid is pCAGGS-FLAG-hUb-WT. |
