Further information: The plasmid was constructed by inserting an EcoRI PCR amplicon, containing a fusion gene between a fragment of the HIV TAT cDNA and the minimal NF-κB inhibiting mouse ABIN-1 cDNA fragment that consists of nucleotides 1446 to 1919 of the coding sequence, flanked by the N-terminal His-tag and the C-terminal E-tag.
The plasmid is useful for highly efficient expression of the fusion gene under the control of the AG promoter and the human CMV-IE enhancer in various mammalian cells.
When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide.
The nucleotide sequence of the mABIN-1 cDNA corresponds with the EMBL Nucleotide Sequence Database accession number AJ242778.1.
Although the depositor described the presence of the Kozak consensus sequence ACCATGG at the start of the fusion gene, the sequence file doesn't contain this sequence, as was confirmed by sequence analysis of this region by BCCM/GeneCorner.
