Further information: The plasmid was constructed by isolating the Myc epitope and His-tag as an ApaI/XmaI fragment from the pCD-MK vector and ligated them in frame to the human MALT1 cds in the pCD-F-HLH-MALT-C464A vector.
The human MALT1 cds contains a C464A mutation.
The plasmid contains a helix-loop-helix domain (HLH) fused to the human MALT1 cds. The HLH domain is a dimerization domain that allows proteins to form homo- and/or heterodimers. The HLH domain consists of the N-terminal sequence of human MALT1 (encoding the first 42 amino acids) and a fragment of the human ets variant gene 6 (ETV6) coding sequence (encoding amino acids 27 to 144).
The parental clone of this vector has been shown to provide resistance to kanamycin, although growth was reduced compared to growth on medium containing ampicillin.
The nucleotide sequence of the wild type human MALT1 cds corresponds with the EMBL Nucleotide Sequence Database accession number AF130356.2 except for a G instead of T in nucleotide position 4, a C instead of A in position 1393, a C instead of G in position 1395, a T instead of A in position 1488 and a G instead of A in position 1950 of the cds nucleotide sequence.
