| Further information: | The plasmid was constructed as follows: the human BCL2 cDNA was PCR amplified and cloned into the unique EcoRI site of pCR II (Dr V. Mironov, Department of Genetics, Ghent University, Belgium). The orientation of the insert was verified by a BamHI digestion. The plasmid contains two selection markers, neomycin (or kanamycin) and ampicillin, both under control of the neo promoter, but each with their own Shine-Dalgarno. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727447.1. The human BCL2 EcoRI fragment of pCAGGShBcl-2 has been sequenced at the Department of Biomedical Molecular Biology (Ghent University, Belgium) and found identical to the DNA sequence described in the Genbank database accession number BC027258.1, except for position 189 of the coding sequence, resulting in a silent mutation. Other name of the plasmid is pCR II (bcl2). |
