Further information: | The plasmid was constructed by inserting C. albicans MET15 genomic DNA as a Sau3AI/EcoRI fragment of YCpCaMET15 between the BglII and BamHI sites of pGAL1PNiST-1. All restriction sites were blunted with T4 DNA polymerase. The orientation of the inserted fragment is antisense relative to the URA3 gene and opposite as to the schematic representation in Viaene et al. (2000). When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727328.1. The nucleotide sequence of the CaMET15 genomic DNA corresponds with the EMBL Nucleotide Sequence Database accession number AF188275.1. The nucleotide sequence of the CaURA3 DNA corresponds with the EMBL Nucleotide Sequence Database accession number X14198.1. HOWEVER, THERE MIGHT BE CORRESPONDENCE WITH AF109400.1: see LMBP 4100. |
