| Further information: | This plasmid gives powerful mammalian expression, also for antisense RNA; inserts are to be cloned by exchanging the stuffer fragment between the XbaI or BstXI sites. Use L929, HeLa, Ch4-2 or COS cells for expression. The BstXI restriction site, present in the human elongation factor 1α promoter, has been removed. When cloning a fragment downstream from the lac promoter it may be advisable to use lacI(q) strains in order to prevent fortuitous expression of a possibly noxious polypeptide. The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727211.1. The nucleotide sequence of this plasmid, originally provided by Dr S. Nagata, has been adjusted to the pCDM8 sequence update. |
