| Further information: | The plasmid was constructed as follows: 1) pGEME6H was cleaved with ApaI (10th nucleotide of the hinge sequence) and blunted (Klenow fragment); 2) an XbaI linker ('TCTAGA') was added by ligation, resulting in the vector pGEME6H3 which contains a termination codon preceding the first Cys codon of the hinge sequence; 3) the XbaI fragment from pGEME6H3 was subsequently deleted by an XbaI digestion. |
