|Cloned DNA:||Mouse anti-hPLAP monoclonal antibody E6(IgG2b,κ) cDNA; truncated heavy chain of the Fab fragment E6F1 (E6Hf1)
|Promoter:||Simian virus 40 early promoter (SV40 early)|
|Terminator:||Simian virus 40 polyadenylation signal (SV40 polyA)|
|Selection marker:||Ampicillin (amp)|
|Replicon:||Escherichia coli plasmid pMB1 origin
Simian virus 40 bidirectional origin (SV40)
|Host range:||Escherichia coli
Mammalian cells; SV40 permissive cells
|Parental clone:||pSV23; pGEME6Hf1|
|Further information:||The plasmid was constructed by inserting the HindIII-XbaI fragment of pGEME6Hf1, containing the cDNA copy of the truncated heavy chain E6Hf1 of the mouse Fab fragment E6F1, into the unique XbaI and HindIII expression sites of pSV23.
This plasmid is similar to pSV23SE6Hf1, but does not contain the SV40 small t-antigen splicing signal.
The translation stops at the beginning of the hinge region and therefore, no cysteine residues involved in the inter-heavy-chain disulfide bonds are present.
The plasmid is designed for transient expression in COS cells or constitutive expression in CHO cells (cotransfection with DHFR plasmids).
