| Further information: | This plasmid was constructed as follows: 1) dC/dG-tailed cDNA of human interferon γ (hIFNG) isolated from SEA-induced human splenocyte cultures was inserted into the unique, filled-in BamHI site of pSV529 in the antisense orientation relative to the SV40 late promoter, resulting in the intermediate construction pSV529HIFNG5; 2) A Sau3AI fragment of this intermediate plasmid, containing the complete hIFNG coding sequence without the dC/dG-tails, was inserted into the unique BamHI site of pSV529 in the sense orientation relative to the SV40 late promoter. This plasmid yields biological active human interferon γ in the supernatant of transfected kidney cells of the African Green Monkey (AP8 cells). The nucleotide sequence of this plasmid corresponds with the EMBL Nucleotide Sequence Database accession number LT727634.1. Other name of the plasmid is pSV529HIFNG5Ss. |
