Further information: The plasmid was constructed by cloning the D. rerio Lrrc4ba extracellular domain coding sequence into the unc5bCD4d3+4-bio vector for Avidity-based Extracellular Interaction Screening (AVEXIS) assays.
This vector expresses the D. rerio Lrrc4ba as a bait protein, linked to a tag containing domains 3 and 4 of the rat CD4 (Cd4d3+4) protein, followed by a biotinylation sequence that is a substrate for the E.coli BirA enzyme and can therefore be monobiotinylated on a specific lysine residue. The Cd4d3+4 tag is recognised by the OX68 monoclonal antibody and is used to quantify the expression of the bait proteins by ELISA. The bait proteins are therefore monomeric and monobiotinylated.
The unc5bCD4d3+4-bio vector was derived from pTT3, obtained from Dr Yves Durocher (National Research Council of Canada, Biotechnology Research Institute).
The nucleotide sequence of the D. rerio Lrrc4ba cDNA corresponds with the EMBL Nucleotide Sequence Database accession number CU468796.1.
