Recipe: Mineral salt sol. (Hutner/Basal Salt) 20 ml

NH4Cl 0.53 mg

KH2PO4 1.4 mg

KNO3 10 mg

MgSO4 x 7H2O 49.3 mg

CaCl2 x 2H2O 14.7 mg

CaCO3 25mg

NaHCO3 25 mg

Peptone (Bacto) 1 g

Yeast extract (Bacto) 1 g

HEPES 2,38 g

Distilled water up to 1L

Adjust pH to 7 with 5 M KOH



For plates: 15 g/l agar (Bacto) 3X washed with distilled water, autoclave separately.



Sterilize 20 min./121°C. After cooling to 60°C add to the medium:

Glucose sol. (25%, sterile-filtered) 4 ml

Vitamin sol. 5 ml

N-Acetylglucosamine 20 ml of 50 g/lstock sol.

Trace element sol.1 ml



Mineral salt sol. (Hutner/Basal Salt):

NTA 10 g

MgSO4 x 7H2O 29,70 g

CaCl2 x 2H2O 3,34 g

Na2MoO4 x 2H2O 12,67 mg

FeSO4 x 7H2O 99 mg

Metall salt sol. "44" 50 ml

Distilled water add to 1L

Dissolve NTA in 700 ml distill water by adjusting pH to 7.2 with KOH. Dissolve all components separately and combine slowly. Sterilize by filtration. Store at 4°C



"Metals 44":

Na-EDTA 250 mg

ZnSO4 x 7H2O 1095 mg

FeSO4 x 7H2O 500 mg

MnSO4 x H2O 154 mg

CuSO4 x 5H2O 39,5 mg

CoCl2 x 6H2O 20,3 mg

Na2B4O7 x 10H2O 17,7 mg

Distilled water 1L

Dissolve the EDTA in 700 ml and add a few drops of concentrated H2SO4 to retard precipitation of the heavy metals if necessary. Store at 4°C. Do not autoclave.



Vitamin solution:

Biotin 4 mg

Folic acid 4 mg

Pyridoxine-HCl 20 mg

Riboflavine 10 mg

Thiamine-HCl 10 mg

Nicotinamide 10 mg

Ca-pantothenate 10 mg

Vitamin B12 0,2 mg

p-Aminobenzoic acid 10 mg

Distilled water 1 L

Dissolve p-Aminobenzoic acid first. Store in the dark and cold



Trace element solution per liter:

Na-Nitrilotriacetat 1500 mg

MnSO4 x H2O 500 mg

FeSO4 x7H2O 100 mg

Co(NO3)2 x 6H20 100 mg

ZnCl 100 mg

NiCl2 x 6H2O 50 mg

H2SeO3 50 mg

CuSO4 x 5H2O 10 mg

ALK(SO4)2 x12H2O 10 mg

H3BO3 10 mg

NaMo04 x 2H2O 10 mg

Na2WO4x 2H2O 10 mg

Sterilize by filtration. Store in the dark and cold
