Recipe: | Peptone 5 g Casitone 5 g Yeast extract 10 g (NH4)2SO4 0,5 g Glucose 5 g Resazurin 1 mg Mineral solution (see below) 40 ml Fatty acid mixture 3,1 ml (see below) Distilled water up to 1 L Mineral solution: CaCl2 x 2H2O 0,3 g MgSO4 x 7H2O 0,48 g K2HPO4 1 g KH2PO4 1 g NaHCO3 10 g NaCl 2 g Distilled water 1 L Fatty acid mixture: Acetic acid 17 ml Propionic acid 6 ml n -Butyric acid 4 ml n -Valeric acid 1 ml iso -Valeric acid 1 ml iso -Butyric acid 1 ml DL-2-Methylbutyric acid 1 ml Dissolve ingredients, boil for 5 minutes, cool to room temperature while gassing with 100% CO2, and add 5 mg haemin (dissolved in 0,5 ml 1N NaOH), 0,2 ml vitamin K1 and 0,5 g cysteine hydrochloride per liter medium. Adjust pH to 6,9 with 8N NaOH while gassing is continued. Switch to 100% N2 gas after pH has reached the desired value, distribute in anaerobic tubes under N2 and sterilize. If needed, add 15 g agar per liter medium. |
