(2.5%, sterile-filtered) 10 ml Vitamin solution 10 ml Adjust pH to 7.5 Artificial sea water : NaCl 23,477 g Na2SO4 3,917 g MgCl2 x 6 H2O 4,981 g CaCl2 1,102 g NaHCO3 192 mg KCl 664 mg KBr 6 mg H3BO3 26 mg SrCl2 24 mg NaF 3 mg Distilled water 1 L Vitamin solution : Vitamin B12 0,1 mg Biotin 2 mg Thiamine-HCl x 2 H2O 5 mg Ca-pantothenate 5 mg Folic acid 2 mg Riboflavine 5 mg Nicotinamide 5 mg Distilled water 1 L. Filter sterilize. Store in the dark and cold (5°C) Hutner’s Basalt salts: Nitrilotriacetic acid (NTA) 10 g MgSO4 x 7 H2O 29,70 g CaCl2 x 2 H2O 3,34 g Na2MoO4 x 2 H2O 12,67 mg FeSO4 x 7 H2O 99 mg “Metals 44" 50 ml Distilled water up to 1 L Dissolve NTA first by neutralizing with KOH (bout 7,3 g), then add other salts. Adjust pH to 6.8 with KOH or H2SO4. Adjust volume to 1 L with distilled water. "Metals 44": Na-EDTA 250 mg ZnSO4 x 7H2O 1095 mg FeSO4 x 7H2O 500 mg MnSO4 x H2O 154 mg CuSO4 x 5H2O 39,5 mg CoCl2 x 6H2O 20,3 mg Na2B4O7 x 10 H2O 17,7 mg Distilled water 1L Dissolve the EDTA in 700 ml, add the heavy metal ions one after the other and add a few drops of concentrated H2SO4 to retard precipitation of the heavy metal ions if necessary. |
