| Recipe: | Yeast extract 0,25 g Mannitol 1,5 g Sodium gluconate 0,5 g KH2PO4 0,5 g MgSO4 x 7H2O 0,4 g NH4Cl 0,4 g NaCl 0,4 g CaCl2 x 2H2O 50 mg Trace element solution 1 ml (see medium 79) Distilled water up to 1 L Adjust pH to 4,9, then add: Ferric citrate solution (0,1 g per 100 ml) 5 ml Biotin solution (2 mg per 100 ml) 1 ml p -Aminobenzoic acid (10 mg per 100 ml) 1 ml Sterilize in screw-capped bottles. Cool to room temperature and add 2 ml of a filter-sterilized 10% Na2S2O3SOlution per liter medium. Incubate in light using a tungsten lamp. If needed, add 15 g agar per liter medium. |
