Solution 5: Microelements



1. Dissolve 1,225 g of MnSO4.4H2O (35) (0,93 g of MnSO4.H2O) in 100 ml pure distilled water

2. Dissolve 0,14 g of ZnSO4.7H2O (74) in 100 ml pure distilled water

3. Dissolve 0,925 g of H3BO3 (125) in 100 ml pure distilled water

4. Mix 1+2+3

5. Weigh 1,1 g of CuSO4.5H2O (20) and dissolve in 50 ml pure distilled water

6. Pipet 5 ml to sol'n 1+2+3

7. Dissolve 0,12 g of Na2MoO4.2H2O (664) in 100 ml pure distilled water

8. When it is completely dissolved, pipet 1 ml to sol'n 1+2+3

9. Dissolve 1,7 g of (NH4)6Mo7O24.4H2O (111) in 100 ml pure distilled water

10. When it is completely dissolved, pipet 1ml in sol'n 1+2+3

11. Adjust the volume of sol'n 1+2+3 to 500 ml



To prepare a 1 liter volume of solid medium

10 ml of solution 1 (macroelements)

10 ml of solution 2 (Ca(NO3)2.4H20)

5 ml of solution 4 (Na Fe EDTA)

1 ml of solution 5 (microelements)

5 ml of solution 3 (vitamins)

10 g of sucrose

Dissolve completely and adjust the volume to 1 liter (H2O Type II)

Adjust the pH to 5.5

Add gelling agent (3 g/l)

Autoclave for 15 minutes at 121°C
