| Further information: | The plasmid was constructed by PCR amplifying the promoter and the terminator of rp2 from T. reesei QM9414 genomic DNA and ligating them into the KpnI-BamHI sites and XbaI-SalI sites respectively of pCAMBIA-1300 (Beijing Ding Guo Changsheng Biotechnology Co.), generating p1300rp2. For heterologus expression of phospho-beta-glucosidase from A. niger, the 2.6-kb BglA gene was inserted into the XbaI site of p1300rp2. The signal peptide coding sequence was included in the amplified BglA fragment. P1300rp2/bgla is useful for heterologous protein expression in T. reesei under control of a constitutive promoter. The nucleotide sequence of the pCAMBIA-1300 vector corresponds with the EMBL Nucleotide Sequence Database accession number AF234296.1. |
