| Further information: | The plasmid was constructed by cloning two homology arms for mouse chromosome 1 into the pCAX-F-hXBP1dDBD-tdTomato vector. The tdTomato coding sequence in this plasmid has been codon optimized for expression in mouse cells. The human XBP1 coding sequence lacks the DNA binding domain (DBD) sequence. The plasmid contains a left homology arm (919 bp) and right homology arm (748 bp) in order to insert the hXBP1dDBD-tdTomato coding sequence in the mouse chromosome 1 (between positions 147.814.420 and 147.814.421), using Tild-CRISPR technology. Other name of the plasmid is pCAX-F-XBP1delDBD TdTomato-TILD-Sense. |
